    Folate-receptor-targeted delivery of doxorubicin using polyethylene glycol-functionalized gold nanoparticles

    Abstract
    Doxorubicin-loaded nanocarriers were produced employing folate-modified polyethylene glycol (PEG)-functionalized gold nanoparticles for targeted delivery to positive folate-receptor cancer cells. Doxorubicin and folate were, respectively, conjugated to activated-folate and activated-PEG. The conjugates formed doxorubicin nanocarrier with an average size of 12 nm in diameter. The drug release response of functionalized gold nanoparticles was characterized by an initial rapid drug release followed by a controlled release.     Abstract
    Doxorubicin (DOX), a common cancer chemotherapeutics, was conjugated to folate-modified thiolated-polyethylene glycol-functionalized gold nanoparticles. The in vitro, controlled release behavior of DOX-loaded gold nanoparticles was observed using porous dialysis membranes (cut-off = 2 kDa). DOX-loaded gold nanoparticles had higher cytotoxicity for folate-receptor-positive cells (KB cells) compared to folate-receptor-negative cells (A549 cells) which were 48 and 62% viable for 10 μM doxorubicin, respectively.     Micro and nanotechnology can potentially revolutionize drug delivery systems. Novel microfluidic systems have been employed for the cell culture applications and drug delivery by micro and nanocarriers. Cells in the microchannels are under static and dynamic flow perfusion of culture media that provides nutrition and removes waste from the cells.     Abstract
    This paper reports the fabrication of electrospun polydimethylsiloxane (PDMS) membranes/scaffolds that are suitable for three-dimensional (3D) cell culture. Through modification the ratio between PDMS and polymethylmethacrylate (PMMA) as carrier polymer, we report the possibility of increasing PDMS weight ratio of up to 6 for electrospinning. Increasing the PDMS content increases the fiber diameter, the pore size, and the hydrophobicity.     In this work, the synthesized graphene oxide/TiO2/doxorubicin (GO/TiO2/DOX) composites were loaded into the chitosan/poly(lactic acid) (PLA) solutions to fabricate the electrospun chitosan/PLA/GO/TiO2/DOX nanofibrous scaffolds via electrospinning process.
