Separation of phenolic glycolipids in mycobacterium bovis BCG by reversed-phase high performance liquid chromatography

    , Article Pakistan Journal of Pharmaceutical Sciences ; Volume 22, Issue 2 , 2009 , Pages 145-149 ; 1011601X (ISSN) Salimi, M ; Pourabdi, L ; Zakeri, M ; Abdipour, N ; Sharif University of Technology
    2009
    Abstract
A crude phenolic glycolipid extract from Mycobacterium bovis BCG was fractionated by column chromatography. A reversed-phase high performance liquid chromatography (HPLC) method with UV detection at 275nm was developed for simultaneous detection and separation of phenolic glycolipids (PGLs) in Mycobacterium bovis BCG. This analysis provides a good resolution. Different solvent systems and columns for HPLC were compared. A system composed of acetonitrile-water in the ratio of 0→80% at a flow rate of 0.8 mL/min and C8 analytical column were found to be optimum for HPLC of the phenolic glycolipids. This simple method is therefore appropriate to purify these compounds present in M. bovis extract...

Multivariate optimization of hydrodistillation-headspace solvent microextraction of thymol and carvacrol from Thymus transcaspicus

    , Article Talanta ; Volume 79, Issue 3 , 2009 , Pages 695-699 ; 00399140 (ISSN) Kiyanpour, V ; Fakhari, A. R ; Alizadeh, R ; Asghari, B ; Jalali Heravi, M ; Sharif University of Technology

    Abstract
In this paper multivariate response surface methodology (RSM) has been used for the optimization of hydrodistillation-headspace solvent microextraction (HD-HSME) of thymol and carvacrol in Thymus transcaspicus. Quantitative determination of compounds of interest was performed simultaneously using gas chromatography coupled with flame ionization detector (GC-FID). Parameters affecting the extraction efficiency were assessed and the optimized values were 5 min, 2 μL and 3 min for the extraction time, micro-drop volume and cooling time after extraction, respectively. The amounts of analyte extracted increased with plant weight. The calibration curves were linear in the ranges of 6.25-81.25 and...

Extraction of fluoxetine from aquatic and urine samples using sodium dodecyl sulfate-coated iron oxide magnetic nanoparticles followed by spectrofluorimetric determination

    , Article Analytica Chimica Acta ; Volume 692, Issue 1-2 , 2011 , Pages 80-84 ; 00032670 (ISSN) Bagheri, H ; Zandi, O ; Aghakhani, A ; Sharif University of Technology
    Abstract
A new method based on the combination of magnetic solid phase extraction (MSPE) and spectrofluorimetric determination was developed for isolation and preconcentration of fluoxetine form aquatic and biological samples using sodium dodecyl sulfate (SDS) coated Fe3O4 nanoparticles (NPs) as a sorbent. The unique properties of Fe3O4 NPs including high surface area and strong magnetism were utilized effectively in the MSPE process. Effect of different parameters influencing the extraction efficiency of fluoxetine including the amount of Fe3O4 and SDS, pH value, sample volume, extraction time, desorption solvent and time were optimized. Under optimized condition, the method was successfully applied...

Optimization of dispersive liquid-liquid microextraction and improvement of detection limit of methyl tert-butyl ether in water with the aid of chemometrics

    , Article Journal of Chromatography A ; Volume 1217, Issue 45 , November , 2010 , Pages 7017-7023 ; 00219673 (ISSN) Karimi, M ; Sereshti, H ; Samadi, S ; Parastar, H ; Sharif University of Technology
    2010
    Abstract
Dispersive liquid-liquid microextraction (DLLME) coupled with gas chromatography-mass spectrometry-selective ion monitoring (GC-MS-SIM) was applied to the determination of methyl tert-butyl ether (MTBE) in water samples. The effect of main parameters affecting the extraction efficiency was studied simultaneously. From selected parameters, volume of extraction solvent, volume of dispersive solvent, and salt concentration were optimized by means of experimental design. The statistical parameters of the derived model were R 2=0.9987 and F=17.83. The optimal conditions were 42.0μL for extraction solvent, 0.30mL for disperser solvent and 5% (w/v) for sodium chloride. The calibration linear range...

Liquid-liquid-liquid microextraction followed by HPLC with UV detection for quantitation of ephedrine in urine

    , Article Journal of Separation Science ; Volume 31, Issue 18 , 2008 , Pages 3212-3217 ; 16159306 (ISSN) Bagheri, H ; Khalilian, F ; Ahangar, L. E ; Sharif University of Technology
    2008
    Abstract
Liquid-liquid-liquid microextraction (LLLME) in combination with HPLC and UV detection has been used as a sensitive method for the determination of ephedrine in urine samples. Extraction process was performed in a homemade total glass vial without using a Teflon ring, usually employed. Ephedrine was first extracted from 3.5 mL of urine sample (pH 12) into a microfilm of toluene/benzene (50:50). The analyte was subsequently back extracted into an acidic microdrop solution (pH 2) suspended in the organic phase. The extract was then injected into the HPLC system directly. An enrichment factor of 137 along with a good sample clean-up was obtained under the optimized conditions. The calibration...
