    Abstract
Fighting with cancer requires the delivery of different therapeutics to the target cancerous cells by taking advantage of the synergistic effects of complementary medicine. Herein, we present a folate-PEGylated niosome as an efficient nanocarrier for targeted co-delivery of hydrophobic letrozole (L) and hydrophilic ascorbic acid (A) to breast cancer cells. The formulation of the niosomal nanocarrier was optimized by varying the ratio of cholesterol and surfactants to maximize the drug loading and minimize the size of nanocarriers. The optimum drug carriers were further functionalized with folate-PEG molecules to enhance the efficiency of drug delivery to the breast cancer cells and prevent...

    Abstract
Context: Elevated expression of the folate receptor (FR) occurs in many human malignancies. Thus, folate targeting is widely utilized in drug delivery purposes specially using nano-radioactive agents. Aims: In this work, we report production and biological evaluation of gallium-67 labeled superparamagnetic iron oxide nanoparticles, embedded by folic acid (67 Ga-SPION-folate) complex especially in tumor-bearing mice for tumor imaging studies. Settings and Design: The structure of SPION-folate was confirmed by X-ray diffraction (XRD), transmission electron microscopy (TEM) and foureir transform infrared spectroscopy (FT-IR) analyses. The radiolabeled SPION-folate formation was confirmed by...

    Abstract
The aim of this work was to synthesize molecularly imprinted polymer-poly ethylene glycol-folic acid (MIP-PEG-FA) nanoparticles for use as a controlled release carrier for targeting delivery of paclitaxel (PTX) to cancer cells. MIP nanoparticles were synthesized by a mini-emulsion polymerization technique and then PEG-FA was conjugated to the surface of nanoparticles. Nanoparticles showed high drug loading and encapsulation efficiency, 15.6 ± 0.8 and 100%, respectively. The imprinting efficiency of MIPs was evaluated by binding experiments in human serum. Good selective binding and recognition were found in MIP nanoparticles. In vitro drug release studies showed that MIP-PEG-FA have a...

    Abstract
Nanoscaled quantum dots (QDs), with unique optical properties have been used for the development of theranostics. Here, InP/ZnS QDs were synthesised and functionalised with folate (QD-FA), D-glucosamine (QD-GA) or both (QD-FA-GA). The bi-functionalised QDs were further conjugated with doxorubicin (QD-FA-GA-DOX). Optimum Indium to fatty acid (In:MA) ratio was 1:3.5. Transmission electron microscopy (TEM) micrographs revealed spherical morphology for the QDs (11 nm). Energy-dispersive spectroscopy (EDS) spectrum confirmed the chemical composition of the QDs. MTT analysis in the OVCAR-3 cells treated with bare QDs, QD-FA, QD-GA, QD-FA-GA and QD-FA-GA-DOX (0.2 mg/mL of QDs) after 24 h indicated...

In vitro release behavior and cytotoxicity of doxorubicin-loaded gold nanoparticles in cancerous cells

    , Article Biotechnology Letters ; Volume 32, Issue 5 , May , 2010 , Pages 649-654 ; 01415492 (ISSN) Asadishad, B ; Vossoughi, M ; Alamzadeh, I ; Sharif University of Technology

    Abstract
Doxorubicin (DOX), a common cancer chemotherapeutics, was conjugated to folate-modified thiolated-polyethylene glycol-functionalized gold nanoparticles. The in vitro, controlled release behavior of DOX-loaded gold nanoparticles was observed using porous dialysis membranes (cut-off = 2 kDa). DOX-loaded gold nanoparticles had higher cytotoxicity for folate-receptor-positive cells (KB cells) compared to folate-receptor-negative cells (A549 cells) which were 48 and 62% viable for 10 μM doxorubicin, respectively. This indicates the potential of these nano-carriers for targeted-delivery. In addition, healthy cell viability was 69% for 10 μM free doxorubicin whereas for the same content of drug in...

    Abstract
In this study, superparamagnetic iron oxide nanoparticles (SPION) embedded by folic acid (SPION-folate) were prepared by a modified co-precipitation method. The structure, size, morphology, magnetic property and relaxivity of the SPION-folate were characterized systematically by means of XRD, VSM, HRSEM and TEM and the interaction between folate and iron oxide (Fe3O 4) was characterized by FT-IR. The particle size was shown to be ≈5-10 nm. To ensure biocompatibility, the interaction of these SPION with mouse connective tissue cells (adhesive) was investigated using an MTT assay. Consequently, gallium-67 labeled nanoparticles ([67Ga]-SPION-folate) were prepared using 67Ga with a high labeling...

Engineering folate-targeting diselenide-containing triblock copolymer as a redox-responsive shell-sheddable micelle for antitumor therapy in vivo

    , Article Acta Biomaterialia ; Volume 76 , 2018 , Pages 239-256 ; 17427061 (ISSN) Behroozi, F ; Abdkhodaie, M. J ; Sadeghi Abandansari, H ; Satarian, L ; Molazem, M ; Al Jamal, K. T ; Baharvand, H ; Sharif University of Technology

    Abstract
The oxidation-reduction (redox)–responsive micelle system is based on a diselenide-containing triblock copolymer, poly(ε-caprolactone)-bis(diselenide-methoxy poly(ethylene glycol)/poly(ethylene glycol)-folate) [PCL-(SeSe-mPEG/PEG-FA)2]. This has helped in the development of tumor-targeted delivery for hydrophobic anticancer drugs. The diselenide bond, as a redox-sensitive linkage, was designed in such a manner that it is located at the hydrophilic–hydrophobic hinge to allow complete collapse of the micelle and thus efficient drug release in redox environments. The amphiphilic block copolymers self-assembled into micelles at concentrations higher than the critical micelle concentration (CMC)...
