Investigating the Effect of Niosomes Containing Simvastatin on Breast Cancer Cell Line

, M.Sc. Thesis Sharif University of Technology Akbarzadeh, Iman (Author) ; Yaghmaei, Soheila (Supervisor) ; Bakhshandeh Abkenar, Haleh (Supervisor) ; Ahangari Cohan, Reza (Supervisor)
    Abstract
Breast cancer is the fifth cause of death from cancer and one of the most common fatal cancers in women. The commonly drugs used to treat cancer often destroy healthy tissues and cells and cause common side effects. It is also necessary to consume a large amount of the drug to achieve a particular effect. One of the applications of nanotechnology is drug delivery. Niosomes are one of the newest drug delivery systems that are nowadays much more widely considered due to better penetration and controlled release. One of the most effective medicines for treating breast cancer is statins. These drugs are a group of lipids lowering drugs that lower the synthesis of cholesterol by inhibiting the...

Simvastatin-loaded nano-niosomes confer cardioprotection against myocardial ischemia/reperfusion injury

    Abstract
Although simvastatin (SIM) has been proven to be a powerful agent against myocardial ischemia/reperfusion (MI/R) injury, poor water solubility, short half-life, and low bioavailability have made it futile while using conventional drug delivery system. Hence, this study aims to investigate therapeutic efficacy of SIM-loaded nano-niosomes on MI/R injury. Surface active agent film hydration method was used to synthesize nano-niosomes. The physicochemical properties of nano-niosomes were characterized using dynamic light scattering (DLS) and transmission electron microscopy (TEM). Moreover, niosomes were characterized in entrapment efficiency (EE) and releasing pattern. Male Wistar rats were...

Modification of well-aligned carbon nanotubes with dihexadecyl hydrogen phosphate: Application to highly sensitive nanomolar detection of simvastatin

    , Article Journal of Applied Electrochemistry ; Volume 44, Issue 2 , February , 2014 , Pages 263-277 ; ISSN: 0021891X Fayazfar, H ; Afshar, A ; Dolati, A ; Ghalkhani, M ; Sharif University of Technology
    Abstract
The first usage of dihexadecyl hydrogen phosphate (DHP)-modified highly oriented multi-walled carbon nanotube (MWCNTs) forests in a sensor configuration was developed to investigate the electrochemical oxidation and determination of simvastatin (SV) in pharmaceutical dosage forms. Synthesis of well-aligned MWCNTs on a conductive Ta substrate by catalytic vapor deposition technique using a common chemical, ethylenediamine, and without being plasma-aided was reported. The electrochemical behavior and oxidation of SV at the aligned MWCNTs/DHP/Ta electrode were discussed in detail through cyclic voltammetry and differential pulse voltammetry. This modified electrode showed considerably higher...

    Abstract
Objective: The objective of this study was to use nano-niosomal formulations to deliver simvastatin as a poor-water soluble drug into breast cancer cells. Significance: Our study focused on the problem associated with poor water-soluble drugs which have significant biological activity in vivo. Methods: Different niosomal formulations of simvastatin were prepared and characterized in terms of morphology, size, encapsulation efficiency (EE), and release kinetic. Antiproliferative activity and the mechanism were assessed by quantitative real-time PCR and flow cytometry. Moreover, confocal microscopy was employed to analyze the cell uptake of simvastatin loaded niosomes to the cancerous cells....

    Abstract
Simvastatin, a well-known antihyperlipidemic drug, has antibacterial activity against a broad range of bacteria, especially Staphylococcus aureus. In present study, a nanoemulsion gel-based formulation containing Simvastatin was developed for infected wound therapy. Therefore, different formulations of Simvastatin nanoemulsion were prepared. Based on droplet size, polydispersity index and zeta potential, the best nanoemulsion formulation containing Simvastatin was selected for development of nanoemulsion gel formulation of drug using carbomer 934 as gelling agent. Thermodynamic stability of Simvastatin nanoemulsion was assessed at different conditions. The in vitro antibacterial activity...

    Abstract
Niosomes, as a kind of drug delivery system, is widely used for the topical delivery of lipophilic drugs. Optimization of niosomes plays an essential role in enhancing their therapeutic efficiencies. This study aims to prepare an optimized niosomal formulation of simvastatin (nSIM), a lipophilic member of statins, through the experiment (Response Surface methodology). Optimized niosomes were characterized in size, polydispersity index (PDI), entrapment efficiency (EE), stability, releasing pattern, and antimicrobial activity. The different molar ratio of surfactant and cholesterol were applied to prepare various formulation of simvastatin loaded niosome. Mean particle size and size...
