  7. Abstract:
  8. Investigation of the behavior of living cells in different biological conditions could play a key role in exploring the main reasons of humans' disorders and diseases. In this regard, many researchers are studying different subjects related to the living cells. Although most of these research activities are based on experimental methods, numerical approach could be utilized to investigate some topics which cannot be studied by experimental methods. In this study a three dimensional model of endothelial cell is proposed for the investigation of cell performance during compression test. The model consists of cell membrane, nucleus and main components of cytoskeleton, namely actin filaments, microtubules and intermediate filaments. Actin binding proteins (ABPs) are also considered as crucial elements in regulating actin cytoskeleton performance. The validity of the model is examined through the comparison of the obtained results with the data presented in previous literature. Not only does the force deformation curve lie within a range of the experimental data, but also the elastic modulus of the cell model is in accordance with former studies.
The existence of ABPs in actin cytoskeleton structure makes the model capable of analyzing internal interactions of cytoskeleton during cell deformation. The obtained results prove that cell and nucleus shape changes are strongly dependant on actin and ABPs concentration. In particular, the augmentation of actin filaments concentration reduces force transmission from cell membrane to the nucleus. It is also indicated that cell stiffening with an increase in applied force is significantly affected by actin filaments reorientation, ABPs reorganization and ABPs augmentation. Besides, the relationship between the elastic modulus of the cell and nucleus, and the relationship between ABPs concentration and cell stiffness are investigated in the present study
