Identification and Characterization Natural Products from Borage Leaves Extract Acting as Anti-amyloid Inhibitors

  8. Natural products obtained from plant extract are well known as a valuable source for research and assessments which eventuates to the discovery of some compounds possessing biological and pharmacological properties. Our aims have been to investigate and identify some natural products having the ability to effect on protein misfolding. In neurodegenerative disorders, like Alzheimer’s and Parkinson’s, a protein or a peptide undergoes conformational changes, and consequently the protein transform to fibrils coined amyloid. To determine the effective anti-amyloid compounds in natural products, Borage leaves was chosen. The plant extract was extracted using soxhlet and applied to hen white egg lysozyme (HWEL) protein which has been used as an amyloid model. For identifying the effective compounds as protein fibrillation inhibitor, the total extract was fractionated by HPLC. After assessment of the whole fractions, one fraction showed a significant effect in reducing the fibril formation which proved by TEM analysis. To understand the mechanism in which this compound works, biochemical assays such as intrinsic fluorescent and ANS binding hydrophobic region, were performed and the conformational change(Amide l) during amyloid formation was also investigated. To identify the nature of the effective compound performed on the isolated compound, Mass spectroscopy, FTIR, 1HNMR, and 13CNMR were done. The results proved that, the effective compound is a derivative of aspidospermine (aspidospermine-17-ol 1, acetyl -19, 21- epoxy -15, 16- dimetoxy ). The results of this research introduced the anti-amyloid property of Borage and an inhibitory effect of the aspidospermine for the first time
