Investigating the Performance of Fluidized bed Dryer in Immobilization and Drying of A-amylase Enzyme

- Enzyme immobilization/drying operations are among the important operations of many biotechnological industrial units. The enzyme used in this study is bacterial alpha-amylase, which is widely used in various industries, including detergents. The production of alpha-amylase enzyme contains a large percentage of water in its composition; therefore, a wet wall tower was designed to conduct the experiments to evaporate the water present in the enzyme liquor, analyze its effects on activity, and also to validate the device. The results showed that in a flow of air around 240m3/hr and ambient temperature of 20◦C, enzyme liquor has a 250% increase in biological activity, which indicates the positive performance of the device. Sodium sulfate and Sodium tripolyphosphate salts have been used as carriers, and two methods have been used to immobilize the enzymatic liquor concentrated, Based on spraying into the carrier. Due to available laboratory facilities, a batch fluidized bed dryer was designed to dry the immobilize enzyme. The function of this dryer was studied to achieve the properties of the product, such as reduction of enzymatic activity and final moisture by changing the process conditions. Designing the test was conducted using advanced design software and response surface method for sensitivity analysis of the temperature of the air entering the bed (30-60 ◦C), the amount of dry matter per outlet air flow rate (0.94 - 8.83 g.hr/m3) and the drying time (5-12min). The results showed the significance of the effect of all three variables, the temperature of the air entering the bed, the drying time and the amount of dry matter on the outlet air flow rate respectively, have the highest effect on the enzymatic activity loss and final moisture. The highest and lowest percentage of enzyme activity loss under different operating conditions during drying were 31.867 and 4.871%, with 0.309 and 4.103% of the final moisture content of the dried material respectively. By optimizing the variables to reach the final moisture content of 1% and minimizing the enzyme activity loss and prediction the model derived by fitting of effective variables, the optimal values for each of the variables of the air temperature to the bed, the amount of dry matter on the outlet air flow rate and the time Drying was obtained to be 30 ° C, 0.94 g.hr/m3 and 10.7 min respectively. The experiment was conducted under the operational conditions and result in 11.021% and 1.275% values for enzymatic activity loss and final moisture respectively
