Extraction and processing optimization of gelatin from fish (otolithes ruber) skin

  4. Gelatin is a denatured protein derived from collagen by thermohydrolysis and has a rheological property of thermo reversible transformation between sol & gel. Gelatin is one of the most important biopolymers, with widespread applications in the food, pharmaceutical, cosmetic and photographic industries. Gelatin is commercially made from skins and skeletons of bovine and porcine and mammalian gelatin has been intensively studied. However, mammalian gelatin is at risk for contamination with bovine spongiform encephalopathy and increase in demand for kosher and halal foods have created a demand for fish gelatin for food applications. Therefore, the objective of this study was to extract gelatin from the skins of Otolithes ruber existing in Persian Gulf, only a few studies have been conducted on warm water fish gelatin and these show that these fish species give gelatin of better functional properties than cold water fish species, and optimization of processing by Taguchi method with Qualitek software. The extraction was carried out by a series of steps involving washing with an aqueous solution of an hydrogen peroxide in order to prevent bacterial degradation of the fish skins and treating the wash skins with an acid. This was followed by a final extraction with hot-water at (55, 60, 65°C) for (4-6-8h), pH between (4-4.5; 4.5-5;5-5.5) and the amount of solution (2.5-3-3.5(VAV)). The result of experiments showed average of performance 12.6% and predicted maximum yields of 24% for gelatin processing at optimum conditions
