Synthesis and Stability Evaluation of Single Enzyme Nanoparticles for Enzymatic Based Processes

, Ph.D. Dissertation Sharif University of Technology Khosravi, Arezoo (Author) ; Vossoughi, Manoochehr (Supervisor) ; Shahrokhian, Saeed (Co-Advisor) ; Alemzadeh, Iran (Co-Advisor)
    Abstract
Enzymes, the nanometric biocatalysts that play a key role in biochemical reactions of living cells and bio-organisms, offer various applications including biosensing, bioremediation and chemical synthesis. A simple bio-conjugation procedure to surround a single horseradish peroxidase (HRP) enzyme molecule with dendritic polyester macromolecules (Polyester-32-hydroxyl-1-carboxyl bis-MPA Dendron, Generation 5) was proposed in this project. The characterization of resultant nanoparticles entitled HRP dendrozyme, was performed by TEM, DLS, GPC and FTIR spectroscopy. The results showed that HRP nanoparticles were spherical in shape and have an average size of 14 nm in diameter. Furthermore,...

Computational Fluid Dynamic (CFD) of Fixed-Bed Reactor for Bio-treatment of Sulfur from Petroleum Cuts

, M.Sc. Thesis Sharif University of Technology kharghanian, Hosein (Author) ; Yaghmaei, Soheila (Supervisor)
    Abstract
Bio-desulfurization is the sulfur removal process from organosulfur compounds by the action of microorganisms (which are capable of removing sulfur). Too many studies have been carried out in the use of biocatalysts for BDS process. In this project, we have used data obtained from a previous experiment in which a trickle bed reactor (diameter: 0.066 m, height: 0.6 m) and a Rhodococcus sp. 12891 starin immobilized on bed particles are used to produce diesel with low sulfur content. Sulfur concentration of inlet diesel is between 200ppm to 540 ppm. parameters of process are recycle Ratio , volumetric flow rate and sulfur content of inlet diesel.
A mathematical model for trickle bed...

Immobilization of Laccase onto Graphene Oxide-based Nano-Composites for Decolorization of Colored Wastewaters

, Ph.D. Dissertation Sharif University of Technology kashefi, Saeed (Author) ; Borghei, Mehdi (Supervisor) ; Mahmoodi, Neyazmohammad (Supervisor)
    Abstract
Using free laccase, enzymatic decolorization of two azo dyes were optimized. The optimum conditions with the aim of maximizing the decolorization of AB92 dye (92.30%) were: dye concentration = 11.85 mg L-1, pH = 5.1, and enzyme concentration = 98.37 mg L-1. While, these conditions to achieve the maximum decolorization yield of DR23 (95.60%) were: dye concentration = 17.68 mg L-1, pH 3.7 and enzyme concentration =97.89 mg L-1.In the second part, the laccase enzyme was covalently immobilized onto GO nanosheets. At the concentration of graphene oxide and laccase enzyme equal to 1 mg mL-1 and 0.9 mg mL-1, respectively, the enzyme loading was 156.5 mg g-1 and the immobilization efficiency was...

Investigating One-pot Three Component Synthesis of ß-Amino Carbonyl Compounds Exploiting Hydrolases: Protein Splicing Enzyme and N-Acetylmuramide Glycanhydrolase

, M.Sc. Thesis Sharif University of Technology Fathali, Yasaman (Author) ; Kalhor, Hamidreza (Supervisor)
    Abstract
The application of biocatalysts in organic reactions refers to the use of enzymes, purified or crude, to increase the reaction rate. The usage of biocatalysts in organic reactions have many advantages, including biocompatibility, ease of separation, high yield, reusability without losing activity, high selectivity, and the use of water as the solvent. Furthermore, the carbon–carbon bond formation is one of the most important reactions in the synthesis of organic compounds, drugs, and biomolecules. Using enzymes in organic reactions can provide insightful information about the enzymes catalytic activities and paves the way for systematic investigation of their mechanisms. The enzymes...
