Design of Drug Nanocarriers Based on Mesoporous Silica Nanoparticles Coated with Smart Polymers

- Mesoporous silica nanoparticles have broad application in drud delivery systems due to their porous structure, functionalization, biocompatibility, high surface area and pore volume. Neverthless, pure mesoporous silica nanoparticles without functionality were not smart material and could not release drug in triggered and controlled manner. For this reason, using smart polymeric coating would be considered. Polymer shells also provide colloidal stability, improved blood circulation lifetime and reduced toxicity which are crucial for efficient in vivo drug delivery. Inflammatory and tumor tissue have low pH and high temperature as compared to health tissue. Therefore, using pH and thermosensitive polymer coating could be effective in triggered drug release. In this research, some pH and thermosensitive polymers were used such as chitosan, poly(caprolactone), poly(acrylic acid), poly(vinyl pyridine), poly(acrylamide), polyglycerol, albumin, dendrimer and polypseudorotaxane. These polymers used as coating on the nanosilica surface and the other magnetite and graphene oxide derivatives. These polymer shells collaped under low pH and high temperature and drug could be released. The use of cylodextrins as gatekeepers and folic acid as targeting ligand has effective role in controlled drug release. The study of cell toxicity assay confirmed enhanced cell uptake and death of cells. Also, increased drug release was seen at cancerous conditions (acidic pH and high themperature) in all polymers which were used as coating of nanoparticles. Moreover, the presence of dendrimer, polypseudorotaxne and human serum albumin in the sturucture of nanocarries provided passive targeting ability. Beside the advantages of these carriers, there are some drawbacks such as non biocompatibility, weak colloidal stability, numerous synthetic routes and low yield.
