- Biodiesel has drawn the attention of many researches to itself as a replacing source for crude oil, recently. Transesterification is one of the most popular methods used for biodiesel production. Enzyme transesterification is a more simple method in comparison with acidic or basic transesterification. However, the high price and low yield of enzyme transesterification has caused many obstacles in its application. Enzyme engineering has gained much attention as a way to improve enzyme performance. In this thesis, biodiesel production by a native enzyme named Bacillus thermocatenulatus, and two engineered-enzymes is studied. First, effective parameters and variables in biodiesel production by native enzyme eas studied and optimized-parameters were determined. Optimized condition of production with 4:1 molar ratio of methanol:soybean oil containing 25 %wt water was determined through three-stage addition of methanol. Second, same procedure was applied for the two engineered enzymes. Effective optimized parameteres for production with 6:1 molar ratio of methanol:soybean oil with 20 wt% of water was determined by addition of methanol 0 and 15 hours after the begining of the reaction. The biodiesel yield of engineered enzyme 1 and 2 was 57% and 35%, respectively. Third, the yield of all three enzymes were studied for products of transesterification reaction of canola,sunflower, and palm oil under optimized condition. Regarding the obtained results, the two engineered enzymes with different amounts of substrate showed a greater yield than the native enzyme
