  7. Abstract:
  8. As the first barrier in front of external damages, skin is prone to the largest number of damages applied to one’s body. Acute wounds are considerably prevalent world-widely, imposing very high costs to governments. The purpose of the current project, is to propose a novel skin scaffold capable of growth factor delivery for enhancing the wound healing process in acute wounds. Such a scaffold should be able to decrease the required time for healing process, also to improve the quality of the regenerated skin compared to available commercial products. Furthermore, it should be highly biocompatible, biodegradable, and non-toxic. In this project, to manufacture the artificial skin scaffold, a methodology based on tissue engineering is employed by proposing a hybrid microsphere/hydrogel system for growth factor delivery. To fabricate the hydrogel substrate a mixture composed of a natural and synthetic biocompatible, biodegradable polymers are employed. The manufacturing method, tensile mechanical properties, viscoelastic properties, degradation rate, biocompatibility, and cell adhesion are assessed in this part. To manufacture the microparticles, the method of water-in-oil-in-water is used. Accordingly, growth factors are entrapped within a polymeric layer. The release profile is also optimized. The investigations are further carried out for the material, microstructure, dimensions, and drug release characteristics. After optimizing both the hydrogel substrate and the microparticles, the hybrid microsphere/hydrogel system is optimized and evaluated in vitro and in vivo. Results of this project can improve the clinical potential of the country and contribute to the country’s healthcare system
