Design, Simulation and Construction of a Microfluidic Device for the Purpose of Target Cells Separation and DNA Extraction

  8. Today, microfluidic experiments have found wide applications in medical sciences, engineering, and chemistry. Because of their small size, microfluidic devices help us to be able to use very little amount of sample for the experiments and also easily control and observe the under-experiment fluid. One of the most important applications of these devices is the separation of cancer cells in blood for counting their number and DNA genetic studies. In the current project we have tried to do the separation of cancer cells from other cells by designing an inertia microchannel. For this purpose, in the first step a geometry for the channel is designed with analyzing the induced forces on the cells. In the next step, for making sure from the separation, a COMSOL simulation is done and then the fabrication is finalized. Finally, experiments are done on the device. Studies of simulations and experiments shows that the designed geometry does the separation with good accuracy. After this, with a process named Lysis which is conducted in a mixer unit in the end of microchannel, cells membrane tears apart and DNA extraction will be done. After providing the necessary conditions, laboratory tests were performed to separate two types of cells, MCF-7 and L929. Tests were repeated three times to ensure results. The average results showed that MCF-7 cells were separated with 61% efficiency and L929 cells with 90.5% efficiency. Also, a test was performed for the focusing process by L929 cells, which its efficiency was 78%. To ensure cell survival during the separation and focusing process, two separate experiments were performed, with a mean survival rate of 94.75%. Finally, in order to investigate the percent of cell separation on the disk, a microchannel on the disk was used, which its efficiency was 64%
