- Idiopathic pulmonary fibrosis is the most common type of pulmonary fibrosis that causes ulcers inside the lungs. In addition to Nintedanib, Pirfenidone is currently the only compound approved by the US Food and Drug Administration in 2014 for the treatment of idiopathic pulmonary fibrosis. Meanwhile, in the pharmaceutical industry, drug synthesis is usually done through batch-wise processes, and pirfenidone is currently only produced through batch-wise processes. Hence, this study aims to change the synthesis method of this drug, which is traditionally performed through batch-wise processes, to a continuous flow process. Therefore, a micro-reactor technology can be adopted which offers a variety of advantages such as better mass and heat transfer, safety, and high surface-to-volume ratio. For this purpose, in this study, a suitable micro-reactor system was first developed by using glass microchips for drug synthesis. Afterward, different experiments were performed at different temperatures, reaction times, and solvents in both the continuous and the batch systems. Following these experiments, the micro-reactor system achieved an efficiency of 31.3% in 40 minutes at a temperature of 170° C, whereas the batch system achieved an efficiency of 18.3% under similar reaction conditions. Moreover, the microreactor device was successful in enhancing the synthesis conditions under further reaction settings.
