- Cancer is the second cause of death around the world. Metastasis is the main reason for cancer patient's death. It has been proven that if metastasis detects in the early stage, the possibility of patients' survival increases. Also, due to the complexity of cancer, there is no unit protocol for the detection and treatment of this disease. Therefore finding a reliable biomarker for diagnosis and treatment application can be very crucial for cancer patients. Circulating tumor cells are one of the common biomarkers for the detection of cancer in its early stages. It has been shown that by isolating and enumerating them, the effectiveness of the treatment approach can be examined. If the number of CTCs in the blood increases, it means the patient does not respond to these drugs or treatment approaches. Also, isolation of viable CTCs and culturing them give us this opportunity to develop a 3D model of patients' tumor for drug testing purposes. One of the biggest challenges in the isolation of CTCs is that they are very rare in the blood; therefore, isolation of them can be very time and labor-consuming. Microfluidic-based methods have recently been introduced for these applications, but most of them use size-based isolation or are not labele-free. In this research, by using the size and electrical properties difference between CTCs and blood cells, the inertial-dielectrophoresis method has emerged to increase the throughput and purity simultaneously. The suggested platform can isolate the MDA-MB-231 cells from blood with a purity of 94 % when the flow rate of the separation process is kept at 650 ul/min
