Comparative Study of Interaction Between Folic Acid (Folate) and Copper(II) Compelexes With a Diimine-Type Ligand

  8. Folic acid is a pterin derivative containing a glutamyl moiety and essential for biological reactions. Pterins are a class of heterocyclic compounds with unique structures and biological functions. Tetrahydrobiopterine has been known to be essential for catalytic hydroxylation of aromatic amino acids by metal ions-containing aromatic amino acid hydroxylases. Metal ions at the active site of these enzymes have been inferred to be closely associated with the pterin cofactors in the catalytic reactions.Information on pterin-metal interaction may give clues to the role of metal ions in enzymatic reactions. With these points in mind, studies have been conducted to verify the formation of ternary complexes containing Cu(II),Folic acid(FA) and 2,2´- Bipyridine (bpy) and 1,10- phenanthroline(phen). Thermodynamic factors such as equilibrium constants (K) for the ternary complexes ,Cu(L)(FA),where L refers to 2,2´- Bipyridine(bpy) and 1,10-phenanthroline(phen), are determined by Uv-Vis spectroscopic method at various temperature in order to quantify chemical equilibria. The stability of the resulting 1:1, Cu(L)(FA) complexes vary in order of [Cu(phen)2]+2> Cu(phen)(Cl)2> Cu(bpy)(Cl)2 showing that the values of K increase as the aromatic ring size increases due to enhancement by noncovalent interaction between coordinated ligands. On the other hand, the absorption spectra of ternary systems differed remarkably, exhibiting a clean isosbestic point at around 385 nm for various complexes throughout the reaction. The temperature dependence of the K can be used for the determination of ΔG, ΔH and ΔS for linear regression fits to the Vants-Hoff equation. There is no derivative from linearity for the plot Ln K v.s 1/T indicating that a 1:1 complex formed for these systems
