Poly (Vinyl Alcohol) Hydrogel/Nano-diamond as a Replacement for the Nucleus Pulposus

  8. The nucleus pulposus, which is a part of intervertebral disc, is normally hydrated and gel-like to maintain normal biomechanical functions of the disc through intradiscal pressures. In a degenerated disc, however, water content of nucleus is significantly reduced, resulting in an abnormal stress state of the annulus. Surgical interventions cannot restore the biomedical function to vertebral segment. One approach to restore the normal biomechanics of an intervertebral disc is to replace the nucleus with a prosthetic material which leads to a restoration of the behavior of the disc. The mechanical behavior of poly(vinyl alcohol) and nanodiamond hydrogel may provide a material suitable for replacement of the nucleus pulposus of the intervertebral disc. In this research, PVA/ND nanocomposite films were fabricated by solution casting method and characterized by XRD, DSC, FTIR, FESEM and mechanical measurements to evaluate the interaction between PVA and ND. It was observed that ND particles were uniformly dispersed within PVA indicating strong interaction through forming hydrogen bonds between hydroxyl groups of PVA and functional groups on ND surfaces. Tensile modulus and strength of film samples were improved remarkably over neat PVA film. Also PVA/ND hydrogels were prepared by gamma irradiation at various doses (15,25,35,45 kGy), and physical properties of hydrogels including gelation, equilibrium water content, and rheological behavior were examined to assess the applicability of these hydrogels for nucleus replacement. Properties were examined as a function of ND concentration as well as gamma dose. By increasing irradiation dose, gel percent and strength of hydrogels increased and their water content was in the range of 80 to 90 wt% similar to the natural nucleus. Results demonstrated that these materials can be proposed as prosthetic materials for replacement of the degenerated nucleus pulposus
