Mechanisms of Conjugated Linoleic acid and Enriched Glycerides Production as Neutraceuticals Food

  8. Dietary CLA production for medicinal purposes needs a safe and selective process. Biological production methods meet this goal. In this study, the microbial species that was suitable for CLA production was selected; then the CLA produced by bioconversion methods was utilized to produce CLA-enriched triglycerides by trans-esterification. Also the biochemical kinetics of CLA production and esterification reactions to produce CLA-enriched triglycerides was studied. In the presence of two substrates, the kinetics of enzymatic reaction was studied. Then the factors influencing on the microbial yield and CLA production including reaction conditions (reaction time, cell concentration and substrate concentration), usage of complementary culture media, concentration of oxygen in the reaction medium, effect of detergent and type of lipase were investigated. Next the parameters affecting on esterification reaction such as amount of enzyme, molar ratio of glycerol and reaction temperature were optimized. The results indicated that Lactobacillus plantarum PTCC1058 showed good ability to produce conjugated linoleic acid, and under optimal conditions (10 mg / ml oil concentration, 15% w/v washed cells and 173 hours) conjugated linoleic acid obtained from ricinoleic acid via bioconversion method, with 2177 mg/l CLA production. Enzymatic transesterification of conjugated linoleic acid showed that lipase inhibitory activity occurs in the presence of alcohol (glycerol, mono- and di-acylglycerol). It was also revealed that the kinetics of enzymatic esterification reaction follows the bi bi ping pong mechanism by dead-end inhibition with acyl receptors. Bio-catalytic reactions were modeled according to the bi bi ping pong mechanism, and the kinetic parameters including Vmax (M / h) =0.328, KCOOH (M) =0.342, KG (M) =0.04526, KiG (M)= 0.329, and R2=0.968 were obtained through the nonlinear regression analysis
