  8. In this thesis, a numerical analysis of micro chamber filling in centrifugal microfluidics is investigated. In the development of micro total analysis systems, it is often necessary to achieve complete and uniform filling of relatively large micro chambers. With centrifugal devices, these large micro chambers must often be orientated perpendicularly to the direction of centrifugal force and are usually bounded by materials with varying surface properties. On the other hand, the mixing characterization of two fluid by centrifugal microfluidic method has been also conducted. The multiphase flow analyses based on Eulerian-Eulerian approach have been conducted and the VOF method was used for modeling of two -phased flow and characterization of fluids interface. The fluent software package is used in order to solve nonlinear PDE equations. Numerical models of the centrifugal PCR micro chamber filling were also developed. The results of simulation have a good agreement with the both experimental data in the literatures and available exact solution methods. The results of simulation indicate that in the micro chamber filling systems, when the fluid reaches to the chambers, due to enlargement of angles a high frequency speed was needed to entry the fluid into the chamber. The results also indicate that, due to the impact of the Euler forces and Coirolis acceleration the velocity profile in the channels and micro chambers have changes to skewed shapes. This effect has a good impact on the uniform mixing of two or more different fluid in the mixing chamber. The mixing characterization in this study shows that the Euler forces that created by changing in rotational velocity of the platform (acceleration/declaration rotational velocity) has a significant impact on uniform mixing of two fluid in the chamber
