Synthesis and Characterization of a Novel Hydrogel Wound Dressing based on Silk Fibroin and Zinc Oxide Nanoparticles

  8. Recently, some wound dressings have been designed that are expected to enhance the wound healing process and prevent blood bleeding and infection. Polymeric dressings have attracted attention due to their biocompatibility, biodegradability, lack of toxicity, and non-allergenic nature. There is widespread interest in using silk fibroin (SF) for this purpose because of its unique properties. Given these points, we have designed an injectable antioxidant hydrogel wound dressing based on SF and Polydopamine(PDA) modified ZnO nanoparticles. First, ZnO nanoparticles were doped with Cu and then modified with PDA (ZnCuO@PDA). XRD, FTIR, SEM, and TEM characterizations were carried out to confirm the accurate synthesis and modification. The ZnCuO@PDA nanoparticles were added to SF solution with different weight ratios and gelation was induced using HRP/H2O2 as crosslinking agent. MTT assay results showed more than 75% cell viability in SF-0.5% ZnO group. Hydrogels had about 40-50% porosity. The biodegradability and water uptake after four weeks were about 20-25% and 300-600%, respectively. Porous structure and cell morphology on hydrogels’ surfaces proved that samples would make a suitable condition for cell adhesion and proliferation due to their interconnected porous structure. Mechanical test results indicated that a higher ZnO fraction in hydrogels led to higher compressive strength and Young’s modulus. Taken together, this system exhibited notable potential as a wound dressing for the treatment of infected wounds
