Scar tissue size following myocardial infarction is an independent predictor of cardiovascular outcomes, yet little is known about factors regulating scar size. We demonstrate that collagen V, a minor constituent of heart scars, regulates the size of heart scars after ischemic injury. Depletion of collagen V led to a paradoxical increase in post-infarction scar size with worsening of heart function. A systems genetics approach across 100 in-bred strains of mice demonstrated that collagen V is a critical driver of postinjury heart function. We show that collagen V deficiency alters the mechanical properties of scar tissue, and altered reciprocal feedback between matrix and cells induces...

Biologic products are large molecules such as proteins, peptides, nucleic acids, etc., which have already produced many new drugs for clinical use in the last decades. Due to the inherent challenges faced by biologics after oral administration (e.g., acidic stomach pH, digestive enzymes, and limited permeation through the gastrointestinal tract), several alternative routes of administration have been investigated to enable sufficient drug absorption into systemic circulation. This review describes the buccal, sublingual, pulmonary, and transdermal routes of administration for biologics with relevant details of the respective barriers. While all these routes avoid transit through the...

Idiopathic pulmonary fibrosis is the most common type of pulmonary fibrosis that causes ulcers inside the lungs. In addition to Nintedanib, Pirfenidone is currently the only compound approved by the US Food and Drug Administration in 2014 for the treatment of idiopathic pulmonary fibrosis. Meanwhile, in the pharmaceutical industry, drug synthesis is usually done through batch-wise processes, and pirfenidone is currently only produced through batch-wise processes. Hence, this study aims to change the synthesis method of this drug, which is traditionally performed through batch-wise processes, to a continuous flow process. Therefore, a micro-reactor technology can be adopted which offers a...
