  7. Abstract:
  8. In the first work, a novel voltammetric biosensor basedon TiO2-nafion-carbon nanoparticles modified glassy carbon electrode (TiO2/N/CNP/GCE) was developed for the determination of DBA. The electrochemical performance of the modified electrode was investigated by means of cyclic voltammetry (CV), different pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. Characterization of the surface morphology and properties of TiO2/N/CNP was carried out by scanning electron microscopy (SEM) and atomic force microscopy (AFM). Effective experimental variables, such as scan rate, pH of the supporting electrolyte, drop size of the cast modifier suspension and accumulation conditions of DBA on the surface of TiO2/N/CNP/GCE were optimized. The best response was resulted at pH 2.0 after accumulation at open circuit for 400 s. Under the optimized conditions, a significant electrochemical improvement was observed toward the electro-oxidation of DBA on the surface of TiO2/N/CNP/GCE relative to the bare GCE. The prepared electrode was used as an approporiate sensor towards DBA determination in the presence of ascorbic acid and uric acid, because of its ability to distinguish betweenoxidation peaks of these compounds, compeletly. Under the optimized conditions, a wide linear dynamic range (6 nM–1 µM) with a low detection limit of 2 nM DBA was resulted. Prepared biosensor showed high sensitivity, stability and good reproducibility in determination of DBA concentrations. Satisfactory results were obtained for DBA analysis of pharmaceutical and clinical preparations using TiO2/N/CNP/GCE. In the second work, the modified glassy carbon electrode by carbon nanoparticles and electrodeposited copper oxide nanoparticles was provided to investigate the electrochemical behavior of nevirapine, under the optimized deposition time and potential (10s, -0.4V). Atomic force microscopy (AFM) was applied for surface characterization of film (CuO-CNP) deposited on the glassy carbon electrode. The optimal conditions were chosen by verification of experimental variables inclouding potential and time accumulation, pH, sweep rate and amount of CNP suspension. Under the omtimized conditions (120s, pH=7.0), the modified electrode shows a significant voltammetric response compared to unmodified electrode. An acceptable dynamic range in three interval (0.1-0.8, 1-10 & 10-100 µM) with a detection limit of 0.066 µM was achieved for the proposed method. It was demonstrated that the reported glassy carbon electrode modified by CuO-CNP is appropriate sensor for the determination of nevirapine in real samples such as human blood serum
