- Abstract:
- In this study, differentiation of rabbit embryonic-derived mesenchymal stem cells to osteogenic cells has been characterized. Bone tissue engineering is based upon the understanding of bone tissue construct and it’s formation in-vivo, and the Preparation of tissue engineered bone constructs to repair large size defects is it’s major goal. We sought to investigate the combined effect of three elemnts of tissue engineering: cells, scaffolds and growth factors. Mesenchymal stem cells are unspecialized cells which due to their unlimited self-renewal capacity and the remarkable ability to differentiate along multiple linage pathways are natural choice for application in tissue repair and reconstruction. Tissues are all composed of matrix and diferrent types of cells. Scaffold also plays a key role in tissue engineering. An important function of the scaffold is to temporarily substitute the extracellular matrix that had assisted the proliferation, differentiation and function of the cells. Growth factors are cytokines which are secreted by some special cells, and act as signaling molecules. In the present study, natural scaffolds prepared from homologous bone tissue which contains necessary growth factors were utilized. Currently applying extracellular matrix requires incorporation of it along with carrier liquid. Using hydrogel form of extracellular matrix decreases the demand of carrier liquids. We have used a perfusion bioreactor to enhance nutrient diffusion. By a simple staining method we have demonstrated applying this bioreactor among with natural hydrogel enabled superior cellular proliferation, spatial distribution and osteogenic induction
