Surface Modification of Super Paramagnetic Iron Oxide Nanoparticles (SPIONs)for Cell Separation

- The aim of this project is the surface modification of super para magnetic iron oxide (SPIONs), leading to oriented covalent bonding of antibody(AB) to these nanoparticles (NPs) and improve the efficiency of cell separation at MACS columns. For this purpose, SPIONs synthesized by co-precipitation method, and the stability of colloidal NPs then was provided by coating with Dextran . We used TGA to measure weight percentage of dextran. Some properties like particle size, hydrodynamic diameter, presence of coating and superparamagnetic properties were characterized by XRD (16 nm), FTIR, DLS (73 nm), TEM (20nm) and VSM (≈76/78 emu/g) method, respectively. Then we tried to immobilize AB on NPs in order to bond them to the head of FC antibodies, this step was studied by FTIR, Ninhydrin - Fusion Assays and UV-Visible .Finally, after conjugation of AB-NPs, we used Flowcytometry to measure conjugated AB concentration and determining the percentage of oriented bonding and efficiency changes. This percentage measured about 80% and cell separation based on free FC was done successfully. The novelty of this project is the surface modifing of SPIONs for higher cell separation efficiency for the first time in Iran and peresenting a new method for oriented conjugation of AB to NPs. Reaserching in this field, by consider the high cost of AB and MACS columns at biological test and existence of limited Iranian company that work at related topics, is limited. This project could be an effective step for the progress of cell separation at nanotechnology in Iran
