Preparation of Gelatin/Pva Porous Scaffolds by Chemical and Gamma Irradiation Methods for Skin Cell Culture

  8. The goal of this research project was to prepare gelatin/PVA porous scaffolds by chemical and gamma irradiation methods to be used for human dermal fibroblast cell culture. First, the polymers were dissolved in hot water and mixed properly to achieve a homogenous solution. The mixture was then frozen at -200C and immersed in ethanol/ glutaraldehyde solution at -200C for 48 hours in order to extract ice crystals and also form chemical crosslinks in gelatin by glutaraldehyde. After being rinsed with distilled water the samples were lyophilized. The effects of polymers solution concentration, gelatin/PVA ratio and glutaraldehyde /gelatin ratio were investigated on characteristics of the scaffolds. Morphology and size of the pores, water absorption capacity, mechanical properties and in vitro biodegradation of the scaffolds were investigated. In irradiation method gamma rays were utilized to form radicals and consequently covalent linkages between PVA and gelatin molecules. In this approach the mixture of PVA and gelatin solutions were exposed to gamma irradiation and after gelation the samples were lyophilized. The effect of polymer concentration, gelatin/PVA ratio and gamma irradiation dose were studied on various properties of scaffolds such as morphology of pores, water absorption capacity, mechanical properties and in vitro biodegradation. The samples made by chemical method showed good properties compared with samples of irradiation method. Consequently fibroblast cells were cultured on three samples of chemical method including different ratios of PVA/gelatin to evaluate the biocompatibility of PVA and cells. MTS assay was conducted to evaluate the influence of PVA on growth and proliferation of cells after 1, 3 and 7 days. There was a slight decrease in cell population with increasing PVA ratio, however all samples showed an increasing trend of cell population with time
