Comparing the Performance of Centrifuge, Membrane Filter and Filter Press for Clarification of Lipase Enzyme

- Lipases (Triacyl Glycerol Acyl Hydrolase, EC.3.1.1.3) are multidimensional biocatalysts which have many applications in food industrials, dairy, fats and oils processing, detergents, synthesis of pure chemical and pharmaceutical materials, paper industrial and cosmetics. Generally, industrial enzymes are produced extracellularly, so their separation from a low-cost complex medium (usually used for production) is one of the cost stages in their production process. Filtration is the most economical method of separation in the production of enzymes, but due to the small size of colloidal particles and cell masses present in the fermentation environment, its use is subject to limitations. In this study, Pseudomonas aeruginosa strain was used to produce maximum lipase extracellular enzyme. The composition of the culture medium included 1gr/lit KH₂PO₄, 3gr/lit (NH₄)₂SO₄, 1gr/lit molasses, 30gr soy oil and 2gr sawdust. The main problem in the separation process is to remove the oil present in this medium, which causes the filters to binding. The optimum pH of the culture medium was 8.2 for maximum lipase production. Filtration operation was performed using centrifugal decanter, centrifugation filtration and plate and frame filter (filter press) with various media. Each of the methods was developed and it was found that the use of Filter press is economically more economical and more efficient in terms of processing. In order to use this equipment, the optimum use of diatomaceous earth filter in its different amounts was obtained 10% by weight and the effects of this soil were investigated in biological separation. The best media for the removal of maximum coarse particles, colloidal particles, cell masses and fats, was cellulose media. Different materials were used for coagulation and clotting of cell masses and colloidal particles, and the best material was obtained by maintaining maximum enzyme activity, sodium hydroxide and optimum consumption of 1% vol. 2 parallel and series filtration were evaluated in the filter press and the series method was selected as the best method. Finally, by this method, the result of the study was a completely transparent liquid with zero absorption capacity inside the spectrophotometer and removal of cell masses and other particles, especially oil. This important result can be proposed for the isolation of other industrial enzymes
