  8. Polymerase chain reaction, abbreviated as PCR, is a method of amplifying the number of copies of a desired DNA sequence through special protocols. The rapid advance of microfluidic devices and the emerging concept of digital microfluidics has resulted into the development of digital droplet PCR (ddPCR) systems with their significant uses in the detection of rare mutations, cancer diagnosis, and surveillance. A large number of micron-sized droplets are required to perform ddPCR. in this study, To investigate the gradient of confinements induced droplet self-breakup mechanism, we carried out the computational fluid dynamics (CFD) simulations for the two-phase flow using a commercial software package ANSYS Fluent. The volume of the fluid model was employed to track the volume fraction of the dispersed and continuous phases (i.e., water and oil, respectively) to capture the droplet breakup process. The inlet flow was set as 2 μl / min and 10 μl / min resulting in droplets with diameters of 118 and 202 μm, respectively. and temperature simulation was performed on the generated droplets and it was found that for the case where the plates are made of glass and Polydimethylsiloxane (PDMS), all the droplets generated are in the allowable range of the PCR thermal cycle
