Optimization of Insulin Release from Beta Cells Using Hybrid Scaffold Bioengineering

  8. Combined composites consisting of nanofibers and hydrogels were fabricated to grow cells in an environment that mimicked the extracellular matrix. Electrospan nanofibers were spun and hydrolyzed under optimized conditions consisting of polycaprolactone (PCL) and gelatin and then incorporated into alginate and gelatin hydrogels. Nanofibril-containing (NF / hydrogel) hydrogel hybrids were ligated by calcium chloride in the presence of encapsulated beta cells. The mechanical properties of NF / hydrogel scaffolds were significantly increased in proportion to the NF content in NF / hydrogels. Beta cells killed in NF / hydrogels showed higher adhesion to the matrix compared to samples without NF. Cells in NF / hydrogels were assayed by changing the increase in NF content for 7 days at concentrations of 5.5 and 15 mM glucose, and by increasing the NF content by about 30% higher level of insulin release than hydrogel scaffolds. Showed. Scaffolds with a concentration of 75 mg / ml NF / hydrogel showed higher levels of survival and insulin release. Thus, NF / hydrogel scaffolds with high concentrations of nanofibrils as a cell culture matrix increase cell-matrix interactions as well as cell survival compared to NF-free hydrogel matrix.
