Design of a Colorimetric Sensor Array Based on Aggregation of Gold Nanoparticles on the Surface of Probiotic Bacteria for Identification and Discrimination of Antiseptic Alcohols

  8. The occurrence of the COVID-19 pandemic and the subsequent rising demand for purchasing alcohol-based sanitizers led to a remarkable prosperity in supplying these hygiene products. Hence, the development of a straightforward and efficient analytical strategy for discrimination of antiseptic alcohols including methanol, ethanol and isopropanol is of great importance. By exploiting the alcohol-sensitive membranes of probiotic bacteria, this research aims to develop a colorimetric sensor array for discriminating methanol, ethanol and isopropanol. In this design, the aggregation of citrate-capped gold nanoparticles (Au NPs) on the thiol-modified surface of two probiotic bacteria, including Lactobacillus Rueteri and Bifidobacterium lactis, was leveraged for fabrication of the sensing elements. The disintegration of the bacterial membrane upon exposure to the antiseptic alcohols inhibits the assembly of AuNPs, thereby precluding the color variations from red to blue. Unequal resistance of the bacteria's membranes against lysis by the alcohols underlies independent response patterns for each analyte. In order to identify and quantify the aforementioned alcohols in sanitizers, the spectral and color response patterns (RGB) of the proposed sensor were examined by linear discriminant analysis and partial least squares regression. The obtained statistical results revealed the outstanding potentiality of the proposed sensor array in identification and discrimination of single-component and multi-component solutions of antiseptic alcohols.
