Simulation and Optimization of α-amylase Enzyme Purification Process
    Using Column Chromatography

, M.Sc. Thesis Sharif University of Technology Rokhforouz, Mohammad-Reza (Author) ; Rosstaazad, Reza (Supervisor)
    Abstract
Enzymes, which are biochemical catalysts, have important applications in different industries such as food, pharmaceutical, medical, chemical identification and etc. Several methods have been used for separation of enzymes among which precipitation, chromatography and two-phase system. In this work, purification of α-amylase has been done by Ammonium Sulfate and Acetone precipitation, gel filtration based on Sephadex, and PEG-salt aqueous two-phase system. Furthermore, mass and momentum conservations’ equations were solved simultaneously to simulate the process based on the finite element method (FEM). Crude enzyme was precipitated with different saturations of ammonium sulfate and the...

Studing the Performance of Rotary Dryer in Drying Enzyme Products

, M.Sc. Thesis Sharif University of Technology Azadi, Mostafa (Author) ; Roosta Azad, Reza (Supervisor)
    Abstract
Drying is one of the important downstream processes in the production of biotechnological products. The dryer design for heat-sensitive materials such as proteins requires special process considerations. In this study, a rotary dryer evaluated for drying protein products. The used liquid enzyme as model, a bacterial α-amylase, first, immobilized on sodium sulfate and sodium tripolyphosphate as carriers in a spraying mode and then, the obtained wet solid materials containing the enzyme, dried in a batch dryer. The performance of this dryer investigated to achieve suitable properties such as the enzyme activity and the final moisture content of product by changing the process conditions....

Determination of Optimum Temperature Profile in Fermentation of B.Subtilis to Maximize Alpha-Amylase Activity

, M.Sc. Thesis Sharif University of Technology Abdollahi Mofrad, Younes (Author) ; Vosoughi, Manouchehr (Supervisor) ; Roostaazad, Reza (Supervisor)
    Abstract
Alpha amylase enzyme is one of the most important enzymes used in a wide range of industries. The enzyme is used in industries such as detergents, food, pharmaceuticals, paper making and textile. Various national and international articles have been published on this enzyme that focused on specific targets. Despite the extensive studies carried out in this area inside the country, there is still no industrial production of alpha amylase and other enzymes in our country. In the view of the fundamental problem of the lack of industrialization of these studies, in addition to the lack of a riskier investor, it seems that there is a lack of industrial vision in these studies. In this study, the...

Design and Operation of Proper Freeze Drying Process to Recover Active Biological Agent

, M.Sc. Thesis Sharif University of Technology Saadat, Mohammad Mahdi (Author) ; Rosta Azad, Reza (Supervisor)
    Abstract
One of the most important unit operations in the production of dry powder biological products involves a freeze dryer apparatus. In this process, Liqueur, which is containing the α-amylase enzyme, undergoes a frigidity condition in which loses its moisture during sublimation. Under these circumstances, applying an adequate supply of sublimation enthalpy without raising the temperature of the frigid material up to melting, plays an important role in the quality of the product and preventing degradation of the α-amylase enzyme. Freeze drying depends on two main factors; time and energy. Furthermore, it is necessary to obtain an optimal temperature and pressure for different steps and...

Studying the Effective Factors and Optimizing Enzyme (α-amylase) Separation in Multi-Enzyme Systems by Agarose Column Chromatography

, Ph.D. Dissertation Sharif University of Technology Ataallahi, Elahe (Author) ; Roosta Azad, Reza (Supervisor) ; Soheila Yaghmaee (Supervisor) ; Yeganeh Sarkandi, Shahin (Co-Supervisor)
    Abstract
In all types of chromatography for the extraction and purification of proteins, a fixed substrate is needed with which the mobile phase containing the proteins interact with them. This fixed bed should contain grains that have characteristics such as sphericity, high surface area, hydrophilicity, binding power to functional groups, chemical stability, low specific absorption, reusability and low cost. Commercially, the most commonly substrates in the ranges of the molecular weight of proteins are agarose and dextran. Agar was extracted from Gracilaria salicornia seaweed and removal of ionic groups was investigated by three methods. In washing with isopropanol, the ionic groups were reduced...

Investigating the Performance of Fluidized bed Dryer in Immobilization and Drying of A-amylase Enzyme

, M.Sc. Thesis Sharif University of Technology Sharifi, Baha (Author) ; Roostaazad, Reza (Supervisor)
    Abstract
Enzyme immobilization/drying operations are among the important operations of many biotechnological industrial units. The enzyme used in this study is bacterial alpha-amylase, which is widely used in various industries, including detergents. The production of alpha-amylase enzyme contains a large percentage of water in its composition; therefore, a wet wall tower was designed to conduct the experiments to evaporate the water present in the enzyme liquor, analyze its effects on activity, and also to validate the device. The results showed that in a flow of air around 240m3/hr and ambient temperature of 20◦C, enzyme liquor has a 250% increase in biological activity, which indicates the...

α-Amylase Immobilization on Nanocomposite Magnetic Silica Particles and Characterization of the Prepared Nanobiocatalysts

, M.Sc. Thesis Sharif University of Technology Sayyahmanesh, Maryam (Author) ; Mashayekhan, Shohreh (Supervisor) ; Arpanaei, Ayyoob (Supervisor)
    Abstract
Enzymes are widely used in industry, however, the expensive cost, low stability, and the limited activity in a certain range of temperature and pH, have limited the usage of such bio-catalists. In addition to increasing and preserving their activity under different conditions, the enzyme immobilization can improve its stability in different pH, temperature values and increase its thermostability, storage stability and the possibility of their recycling. Meanwhile, utilizing the super-magnetic systems could be very helpful as well. For instance, separation of the the magnetic nano-particles like Fe3O4 from reaction system using an external magnetic field is easily possible. In this study,...

studying the Effect of Adding Nanoparticles in Polymer/salt Aqueous Two Phase Systems for Separating of Alpha-amylase Enzyme

, Ph.D. Dissertation Sharif University of Technology Dehnavi, Mohsen (Author) ; Vossoughi, Manoochehr (Supervisor) ; Pazuki, Gholamreza (Supervisor)
    Abstract
The effects of adding nanoparticles in partitioning of α-amylase in polymer/salt aqueous two phase system (ATPS) was studied in this research. First the binodal curves of magnesium sulfate salt with PEGs 2000,3000,4000,6000 and 10000 at 23 ° C has been plotted and the suitable system which was PEG 6000/ magnesium sulfate has been selected for studying the partitioning of enzyme. Thereupon the partition coefficient of α-amylase was measured and then effect of adding silica and graphene oxide nanoparticles was investigated. It was observed that nanoparticles caused a dramatic increase in partition coefficient and yield recovery (of top phase). For further investigation the partitioning of...
