Mathematical Modeling of 188Re-PSMA in Rats and its Human Absorbed dose Estimation

  8. Prostate cancer is the second most common cancer in men and the fourth leading cause of death in the world. In this study, the 188Re-HYNIC-PSMA radiolabeled compound was produced as a compound for use in the treatment of prostate cancer and its metastases under optimal labeling conditions and its radiochemical purity was obtained above 98% using RTLC and HPLC chromatographic methods. This compound is stable at room temperature and in human blood serum for at least 48 hours after preparation. The biological distribution of this compound was investigated in each rat tissue and the amount of injected activity per unit mass of each tissue (%ID/g) was determined. The bio-distribution of 188 Re-HYNIC-PSMA labeled compound was compared with the bio-distribution of free rhenium, which showed significant differences in the tissue uptake of the two group. This difference is well seen in prostate tissue that has a prostate-specific membrane antigen receptor. Bio-distribution modeling was performed by compartmental method based on animal data using ANACOMPTM code and information obtained from ICRP-80. Dosimetric calculations of different human organs were performed based on MIRD method and through extrapolation of animal data for humans using modeling results. The results of the distribution of 188Re-HYNIC-PSMA labeled compound in healthy rats showed that the highest activity accumulates in the kidneys, as the main route of elimination of radioactivity from the body, and the highest dose in human tissues is related to kidneys and is equal to 0.69± 0/08 mSv / MBq. These data are in complete agreement with published animal and human studies for other PSMA compounds. In general, the results showed the possibility of producing 188Re-HYNIC-PSMA labeled compound, with high specific activity with the available facilities in the country. Preliminary studies of this labeled compound also demonstrated that this compound has a high potential for use in the treatment of prostate cancer
