  7. Abstract:
  8. Liver metastasis is the leading cause of death in patients with colorectal cancer (CRC). An increasing number of studies have shown that mRNAs, miRNAs, can play an important role in various biological processes that are related to cancer. Identifying and determining the relationship between them can help to diagnose and treat cancer. The aim of this study is to identify miRNA and molecular mRNA markers in the stage of polyp formation in order to diagnose colorectal cancer early and also to introduce therapeutic targets in the stage of metastasis. In this study, two microarray data sets GPL15207 and GPL16384 were analyzed to identify different miRNAs and mRNAs from the GEO database in three stages of normal tissue, polyp tissue, and tissue in the liver metastasis stage of cancer patients. At first, the differential analysis of mRNA data and significant miRNAs from the EnrichR database was performed and the biological pathway related to it was analyzed. In the end, the diagnostic indicators and treatment targets were prioritized based on the number of repetition in the concepts of centrality.Based on the results, in the stage of polypization, VEGFA، TIMP1 ، SPP1 mRNAs and hsa-mir-378a-3p,hsa-mir-105,hsa-mir-373-3p ,hsa-mir-503-5p miRNA were proposed as the most significant diagnostic markers, and in the stage of metastasis, TTR , SERPINC1 and AHSG mRNAs were proposed as the most important therapeutic targets. In the polyp stage, microRNAs hsa-mir-373-3p, hsa-mir-378-3p and hsa-mir-105 were introduced as new markers that regulate VEGFA and in the metastasis stage AHSG were introduced as a new target treatment which requires a practical experiment in the laboratory. In general, the network is an important method for identifying indicators and treatment methods.
