Preparation of Nanoparticles Based on Hyperbranched Polyglycerol as Anti Cancer Drug Delivery Systems

, M.Sc. Thesis Sharif University of Technology Yazdi, Mojtaba (Author) ; Pourjavadi, Ali (Supervisor) ; Adeli, Mohsen (Co-Advisor)
    Abstract
Successful formulation development for poor water soluble drugs is a critical, long-standing and challengeable problem in cancer treatment. One of the methods for transporting low-soluble drugs is using nanoparticles which are able to pass through capillary walls of tumor tissues due to their small size. An example of the nanoparticles is micelle systems. Functionalized β-cyclodextrin (β-CD) by hyper branched polyglycerol (HPG) can dissolve low-soluble compounds. This polymer can be used in preparing micelles. In this project, β-cyclodextrin (β-CD) by anionic reactions is initiator for polymerization of glycidol monomer. Hydrophobic cavity of the β-cyclodextrin (β-CD) in placing the phenyl...

, M.Sc. Thesis Sharif University of Technology Gholami, Mahsa (Author) ; Vossoughi, Manouchehr (Supervisor) ; Kazemi, Akhtarolmolouk (Supervisor)
    Abstract
Paclitaxel is used as a drug against breast and ovarian cancers. Although paclitaxel has high biologic activity, it has very pour solubility in water like some other drugs. Since drugs should be solved in water to be absorbed by the body, for absorb enhancement, we should use carriers which includes both polar and non polar sites. eta-Cyclodextrin which has this feature is capable for producing inclusion complex in its hydrophobic pores with non polar materials. In this project by adding polymeric polyacrylamide branches on beta-Cyclodextrin, polymeric nano structure has been developed to enhance water solubility in comparison with pure beta-Cyclodextrin. Study on producing polymeric nano...

, Ph.D. Dissertation Sharif University of Technology Zarrabi, Ali (Author) ; Vossoughi, Manouchehr (Supervisor) ; Adeli, Mohsen (Supervisor) ; Shokrgozar, Mohammad Ali (Supervisor)
    Abstract
Today, human is facing and struggling with one of the most deadly disease, Cancer. Accordingly, several research studies are aimed to develop new anticancer drugs. Among the prescribed drugs are Paclitaxel. Unfortunately, despite the anticancer activity, paclitaxel has very low water solubility. To solve this problem several alternatives have been proposed by researchers, e.g. using co-solvents of ethanol:Cremophor El®, as is the case in commercial drug, Taxol®. Unfortunately, these co-solvents have some dangerous and sometimes mortal side effects. To reduce its side effects, researchers have proposed using cyclodextrin as a carrier for paclitaxel on the basis that cyclodextrin has a...

, M.Sc. Thesis Sharif University of Technology Amin, Shiva Sadat (Author) ; Pourjavadi, Ali (Supervisor)
    Abstract
One of the major problems in cancer treatment is the side effects of the drugs. Nowadays, scientists are developing smart nanocarriers which used for diagnosis and delivering drugs in order to circulate through blood vessels, pass the immune system, attach to cancer cells and kill them without any side effects. In the first research of this thesis, magnetic nanoparticles were coated by modified alginate and used as smart nanocarriers. Magnetic nanocarrier were sysnthesized based on hydrophobic coating of oleic acid and hydrazine oleate-modified alginate shell. The resulting carrier is pH-sensitive and the alginate shell removes in the low pH medium. DOX was placed between hydrophobic chains....

, M.Sc. Thesis Sharif University of Technology Asadi, Farshid (Author) ; Saifkordi, Aliakbar (Supervisor) ; Vossoughi, Manouchehr (Supervisor)
    Abstract
Some widely used drugs such as paclitaxel (PTX) are poorly water soluble. The clinical use of PTX with solubilizer causes side effects. These problems limited its application in treatments. In this work has been studied the effect of β-CD-g-PG (HPCD) hybrid nanomaterials, conjugated PG branches onto β-CD core, on the water solubility of PTX. First HPCD has been synthesized. Then the inclusion complex of PTX with HPCD was prepared. The inclusion complex efficiency for PTX:HPCD complex in 1:25 mole ratio was 87%. Then solubility of PTX with HPCD in water has been determined, showing Ap type phase solubility diagram. Then stability constants were calculated. The first order and second order...

    Abstract
The synthesis of a new drug delivery system based on hybrid nanomaterials containing a β-CD core and hyperbranched PG is described. Conjugating PG branches onto β-CD not only increases its water solubility but also affects its host/guest properties deeply. It can form molecular inclusion complexes with small hydrophobic guest molecules such as ferrocene or FITC with reasonable release. In addition, the achievable payloads are significantly higher as for carriers such as hyperbranched PGs. Short-term in vitro cytotoxicity and hemocompatibility tests on L929 cell lines show that the hybrid nanomaterial is highly biocompatible. Due to their outstanding properties, β-CD-g-PG hybrid nanomaterials...

Paclitaxel/β-CD-g-PG inclusion complex: An insight into complexation thermodynamics and guest solubility

    , Article Journal of Molecular Liquids ; Volume 208 , August , 2015 , Pages 145-150 ; 01677322 (ISSN) Zarrabi, A ; Vossoughi, M ; Sharif University of Technology

    Abstract
Paclitaxel as one of the most effective anticancer drugs has low aqueous solubility. This inevitably reveals its commercial formulation in Cremophor EL®/ethanol mixture. However, this formulation leads to several severe side effects such as hypersensitivity reactions, neurotoxicity and nephrotoxicity. Inclusion complexation has been introduced as a practical approach in increasing paclitaxel aqueous solubility. To this end, a hybrid nanocarrier system based on hyperbranched polyglycerol and β-cyclodextrin is designed with key components uniquely structured at nanoscale and evaluated according to medical requirements. Paclitaxel is included in the hydrophobic cavity of cyclodextrin as guest...

Structure and conformation of α-, β- and γ-cyclodextrin in solution: theoretical approaches and experimental validation

    , Article Carbohydrate Polymers ; Volume 78, Issue 1 , 2009 , Pages 10-15 ; 01448617 (ISSN) Tafazzoli, M ; Ghiasi, M ; Sharif University of Technology

    Abstract
The anomeric carbon chemical shifts of α-, β- and γ-cyclodextrin in solution were studied experimentally and theoretically by NMR and two-layer ONIOM2 (B3LYP/6-31G*-GIAO: HF/6-31G*-GIAO) variant. The dependence between the anomeric carbon chemical shift and the glycosidic bond φ and ψ dihedral angles in d-Glcp-d-Glcp disaccharides with and (1 → 4) linkages in α-configurations were computed by Gauge-Including Atomic Orbital (GIAO) ab initio and ONIOM in water solvent using PCM methods. Complete chemical shift surfaces versus φ and ψ for this disaccharide were computed. We also present empirical formulas of the form 13C δ = f(φ, ψ) obtained by fitting the ab initio data to trigonometric series...

    Abstract
In this research a novel controlled anticancer drug delivery system with dual pH and thermal responses was designed based on magnetic mesoporous silica nanoparticles that were anchored by β-cyclodextrin and coated by poly(N-isopropylacrylamide) (PNIPAM). Results demonstrated that the behavior of doxorubicin (anticancer drug) release depended on pH and temperature conditions. At endosomal pH (pH 5.5) the amount of drug release enhanced because the cap was removed from the pores. Furthermore, PNIPAM shell collapsed above the lower critical solution temperature and the releasing of drug increased. Thus, this nanocarrier would have the potential to be applied in the tumor therapy. © 2017 Taylor...

    Abstract
A noncovalent functionalization of the edges of reduced graphene oxide (RGO) with β-cyclodextrin-graft-hyperbranched polyglycerol (β-CD-g-HPG) was successfully performed via a host-guest interaction. The results showed that β-CD-g-HPG disperses the graphene sheets better than pure β-CD or HPG. The resulted supramolecular structure is stable in neutral water medium more than one week. However, in acidic medium the host-guest interaction is collapsed and graphene nanosheets precipitate. © 2017, Copyright © Taylor & Francis

    Abstract
Superparamagnetic Iron Oxide Nanoparticles (SPIONs) were synthesized and coated with pseudopolyrotaxanes (PPRs) and proposed as a novel hybrid nanostructure for medical imaging and drug delivery. PPRs were prepared by addition of α-cyclodextrin rings to functionalized polyethylene glycol (PEG) chain with hydrophobic triazine end-groups. Non-covalent interactions between SPIONs and PPRs led to the assembly of SPIONs@PRs hybrid nanomaterials. Measurements of the 1H Nuclear Magnetic Resonance (NMR) relaxation times T1 and T2 allowed us to determine the NMR dispersion profiles. Comparison between our SPIONs@PRs hybrid nano-compound and the commercial SPION compound, Endorem®, showed a higher...

    Abstract
Chrysin is a bioflavonoid which possesses a wide range of important biological activities. In present study, we used a quantum mechanical approach to shed light on the antioxidant ability and antioxidant mechanism of chrysin to scavenge hydroxyl radical (˙OH) in solution phase. The analysis of the theoretical bond dissociation enthalpy (BDE) values and spin density of the radicals to determine the delocalization possibilities at B3LYP/6-311++G** level clearly shows the importance of the A-ring and the 7-OH group in antioxidant reactivity. In the next step, the inclusion of chrysin with β-CD has been investigated extensively using theoretical methods. Density functional theory (M05-2X)...
