In silico design of novel anticancer drugs with amino acid and carbohydrate building blocks to inhibit PIM kinases

    , Article Molecular Simulation ; Volume 48, Issue 6 , 2022 , Pages 526-540 ; 08927022 (ISSN) Kalhor, S ; Fattahi, A ; Sharif University of Technology

    Abstract
PIM-1 is a serine-threonine kinase mainly expressed in tissues like the Thymus, spleen, bone marrow, and liver. Overexpression of PIM kinases occurs in various types of human tumours, such as lymphomas, prostate cancer, and oral cancer. As a result, the design of drugs to inhibit PIM-1 in cancerous cells has attracted much attention in recent years. This study aimed to design the alternative inhibitors for PIM-1 kinase, which are based on carbohydrates and amino acids and are expected to be non-toxic with the same chemotherapeutic effects as the traditional known anticancer drugs. The combinatorial use of quantum mechanics (QM) and molecular dynamic simulation (MD) has enabled us to...

    Niosomal formulation for antibacterial applications

    Abstract
Infection is a disease that is mainly caused by different Gram-negative and Gram-positive bacteria. Treatment of infections requires a considerable amount of antibiotics, which can cause serious damage to the patient's body. Delivering the antibiotic only to the site of infection can prevent these destructive effects, such as the destruction of the normal intestinal flora. The drug delivery system through carriers will take antibiotics into a part of the body involved in the disease. Niosome nanoparticles, which have been made from non-ionic surfactants, have been emerging as ideal drug/antibiotics delivery vehicles. Recently, niosome formulations have been targeted to reduce toxicity and...

Development of HAp/GO/Ag coating on 316 LVM implant for medical applications

, Article Journal of the Mechanical Behavior of Biomedical Materials ; Volume 126 , 2022 ; 17516161 (ISSN) Ahmadi, R ; Izanloo, S ; Sharif University of Technology
    Elsevier Ltd  2022
    Abstract
In this study, antibacterial activity, biocompatibility, and corrosion resistance of 316 LVM implants were improved using the development of HAp/GO/Ag nanocomposite coatings by the dip-coating method. The XRD and FTIR results confirmed the synthesis of HAp/GO/Ag nanocomposites. HAp/Ag nanoparticles (68 nm) bound to epoxy, hydroxyl, and carboxyl functional groups on GO sheets (size of GO sheets varies from 255 to 1480 nm) by electrostatic interaction. FESEM images showed that HAp/GO/Ag coatings had higher density and fewer micro-cracks than pure HAp coatings. In addition, HAp/GO/Ag coatings showed optimized nano-hardness (4.5 GPa) and elasticity modulus (123 GPa). The results of...

A multi-objective approach to optimize the weight and stress of the locking plates using finite element modeling

    , Article Proceedings of the Institution of Mechanical Engineers, Part H: Journal of Engineering in Medicine ; Volume 236, Issue 2 , 2022 , Pages 188-198 ; 09544119 (ISSN) Rafiei, S ; Nourani, A ; Chizari, M ; Sharif University of Technology
    SAGE Publications Ltd  2022
    Abstract
This paper aims to identify an optimum bone fracture stabilizer. For this purpose, three design variables including the ratio of the screw diameter to the plate width at three levels, the ratio of the plate thickness to the plate width at three levels, and the diameter of the bone at two levels were selected for analysis. Eighteen 3D verified finite element models were developed to examine the effects of these parameters on the weight, maximum displacement and maximum von Mises stress of the fixation structure. Considering the relations between the inputs and outputs using multivariate regression, a genetic algorithm was used to find the optimal choices. Results showed that the diameter of...

Efficient seismic risk assessment of irregular steel-framed buildings through endurance time analysis of consistent fish-bone model

    , Article Structural Design of Tall and Special Buildings ; Volume 31, Issue 2 , 2022 ; 15417794 (ISSN) Ahmadie Amiri, H ; Hosseini, M ; E. Estekanchi, H ; Sharif University of Technology

    Abstract
The seismic risk framework of building structures has been presented to reduce earthquake-induced adverse consequences. In this context, probabilistic analysis of engineering demand parameters (EDPs) is always associated with many uncertainties and high computational demand for use in practical applications. This study has been presented to efficiently estimate the distribution of EDPs in probabilistic seismic risk assessment of irregular steel moment-resisting frames (SMRFs). For this purpose, the incorporation of the consistent fish-bone (CFB) model and the endurance time (ET) analysis method has been used. The proposed method (i.e., CFB-ET) has a substantial impact on reducing the...

    Na Alginate/PVP/Hap nano-composites Hydrogels

, M.Sc. Thesis Sharif University of Technology Kamali Moghadam, Zahra (Author) ; Frounchi, Masoud (Supervisor)
    Abstract
In this research, we developed a drug release system of two biocompatible and biodegradable polymers and biocompatible ceramic nanoparticles. Hydrogels of sodium alginate (SA) were crosslinked using calcium chloride. The SA hydrogels were blended with polyvinyl pyrrolidone (PVP) and mixed with hydroxyapatite nanoparticles (HAP) to make hydrogel nanocomposites in the form of microbeads as drug carriers. Ciprofloxacin was selected as a model antibiotic drug for treatment of bone infection. It was found that SA and PVP form hydrogen bonds and are miscible at whole range of concentrations. Indeed, the SA/PVP blends may be considered as interpenetrating polymer networks (IPNs). HAP nanoparticles...
