- Natural tissues of body consist of Extra Cellular Matrix (ECM) and cells. Scaffold produces a temporary matrix for cells and facilitates oxygen and nutrients transport which enhances tissue regeneration. Therefore, attempts in regeneration of tissues and organs by scaffolds attracted great interest in today’s researchs. Choosing scaffold fabrication method and its polymer type, play a key role in scaffold specifications as they determine scaffold properties. In this project electrospinning was chosen as the scaffold fabrication method which is because of the unique properties of scaffolds fabricated by this method and also its ability in producing micro and nano fibers. At first set of experiments in this research, only one of the Polycaprolactone or Gelatin were used in scaffold fabrication as synthesis and natural polymer; then in the next runs, they were used together by “mixing” and then “layer by layer” electrospinning method. Nano Hydroxyapatite and Titanium Oxide were added to polycaprolactone scaffold which resulted in better wettability, bioactivity and mechanical properties of scaffold. Appropriate properties of 3-layer scaffold and its ability to mimic ECM which is due to its 3 dimensional structure resulted in choosing this scaffold and adding nanoparticles to it. This scaffold showed great wettability and bioactivity and extraordinary great mechanical properties compared to the other tested methods in this research and also all other available research papers solving the strength problem of electrospinning scaffolds
