- Tumor markers are substances that are naturally found in blood and other fluids in body, but in abnormal conditions such as cancer and metastasis, their level increase. The aim of researching about tumor markers is to find a way for discovering cancer only with a blood test and replace with the current methods such as ELISA, and any type of cancer can be detected by these tests. In this study, molecular imprinted polymers were synthesized with biomolecules. To do that, silica microparticles were used as solid base and after functionalizing their surface, a thin film of polymer was synthesized on them. To optimize the components concentrations and the polymer thickness on the silica surface, design of experiments was done by Taguchi method and optimum conditions for polymer imprinting with bovine serum albumin were obtained. After polymer synthesizing, the appropriate pH for rebinding of the template molecule was investigated and pH 7 was selected for the most rebinding quantity. Template molecule rebinding in molecular imprinted polymer and non imprinted polymer was evaluated and imprinting factor was calculated. The rebinding performance of MIP for the template molecule in solutions with different initial concentrations was also studied, and maximum initial concentration obtained was 1 mg/ml. Afterwards, molecular imprinting with CA15-3 antigen with optimum conditions obtained for BSA, was performed. In the case of industrialization, this method is easier and less expensive than current methods and is resistant in differnet environmental conditions such as temperature, pressure and pH
