Modifying Collagen Membrane Used for Guided Tissue Regeneration

- In this study, lyophilized advance platelet rich fibrin (A-PRF) was used in combination with collagen membrane for the first time to accelerate bone regeneration as a result of growth factor release. Firstly, collagen was extracted from calf skin and analyzed using SDS-PAGE. FTIR has shown that EDC/NHS were chemically crosslinked collagen to collagen and collagen to chitosan. The morphology of collagen, collagen/chitosan composite membrane with different chitosan content, lyophilized A-PRF attached to membrane were showed using SEM images. The pore sizes were varied from 100 to 300 µm. Response surface methodology (RSM) was used to design experimental condition and to correlate the effect of parameters, including chitosan/collagen weight ratio (chit/col) and A-PRF concentration on young modulus, mesenchymal stem cell proliferation and degradation rate of the membrane. Results showed that young modulus of the membranes were intensified by increasing chitosan/collagen weight ratio and decreasing A-PRF concentration from 3 Mpa to 8 MPa. Cell proliferation of membrane was intensified by both increasing chit/col and A-PRF concentration. Additionally, results showed that as chitosan/collagen weight ratio increased and A-PRF concentration decreased, degradation rate of membrane decreased from 90 to 20 percent after 4 weeks. Finally, based on Design Expert Software calculation for minimizing degradation rate and maximizing both young modulus and cell proliferation rate, chit/col 4 and A-PRF concentration 0.58 (mg/ml) was suggested for membrane fabrication
