Design of Injectable Hydrogel Scaffold based on Smart Polymer in Tissue Engineering

  8. Nerve damage is one of the factors affecting the quality of life of patients. The nervous system does not have the ability to repair large injuries, and autologous transplantation, which is the standard treatment method for nerve injuries, is faced with a shortage of donors and a decrease in the function of the donor site. Tissue engineering hydrogels, due to their similarity to the natural tissue of the stomatal body, are a hope for the repair of nerve tissue. In this research, an injectable, minimally invasive and temperature-sensitive hydrogel based on chitosan-etaglycerophosphate-sodium hydrogen carbonate salt or trisodium phosphate salt containing aligned nanofibers made of gelatin and polyvinyl alcohol conducted by carbon nanotubes, in order to physically guide axons and electrical cell-to-cell communication was synthesized. The Young's modulus of the hydrogel was in the range of the Young's modulus of the nervous system (0.002-0.0917 MPa). In samples with low Young's modulus, the addition of nanofibers increased the Young's modulus by 23 times and reached the Young's modulus of the spinal cord. Also, the initial biodegradability rate was equal to 50% and then this rate slowed down, as a result of which it not only did not prevent the growth and proliferation of new tissue, but considering the long-term repair of the nervous system, it can play a supporting role for it. Samples containing 0.04 grams of sodium hydrogen carbonate salt and 0.02 grams of sodium phosphate salt were selected as optimal samples. According to the results of the cell viability test, these scaffolds are not toxic and due to the creation of cell adhesion sites by gelatin and the conductivity of the fibers, it causes the growth and proliferation of nerve cells compared to samples containing insulating nanofibers.
