HAP/Agar Nanocarriers for Bone Anti-infection Drug

  8. Successful treatment of bone infections is a major orthopedic challenge due to the physiological and anatomical features of bone. In this project, we prepared a drug delivery system with injectable and biodegradable polysaccharide agar containing alginate, polyvinyl alcohol and hydroxyapatite nanocarriers composite microspheres. Alginate and polyvinyl alcohol composite microspheres are crosslinked by calcium and freeze-thawing technique. Using the freeze thawing process for polyvinyl alcohol crosslinking, in addition to alginate crosslinking, improved hydrogel swelling behavior and enhanced drug loading and thus slowed drug release, drug loading increased with increasing PVA percentage from to and % from % to and 45%. Agar substrate is for the facility of injection. In this project, the effect of polyvinyl alcohol and different percentages of nano-hydroxyapatite on the release of ciprofloxacin as a model drug in saline phosphate buffer with pH = . has been investigated, drug release decreased from 5% to % and % by increasing the percentage of PVA from to % and % of drug release in the first hours. Ultraviolet–Visible (UV–Vis) spectroscopy was used to obtain the concentrations of the released drug and adsorbed drug on the nanoparticles. Importantly, drug loading on hydroxyapatite nanoparticles shows a sustained release profile. Characterization of microspheres / nanocarriers was performed by infrared spectroscopy, X-ray diffraction, and scanning electron microscopy of the samples. the antibacterial activities of composite hydrogels were demonstrated against Gram-negative E.coli bacteria and kinetics of the systems were investigated by two models, Higuchi and Korsmeyer-peppas
